Activities
Year 1
- Generation of three cDNA libraries from infected cells (representing three different temporal phases of infection).
- Performing positive selection for cDNA clones harbouring viral genes.
- Generation of glycerol stocks and plasmid DNA stocks for each viral cDNA clone (Parts a, b and c will be performed at the University of Rijeka).
- Sequence analysis of viral clones at OSU. The rationale for this being done at OSU is that it can interact face-to-face with the sequencing facility to advance the specific needs and rapidly re-submit samples for sequencing at little or no cost.
- Database construction.
- Microarray platform selection.
Year 2
- Completion of sequencing of cDNA libraries generated in year 1 at OSU.
- Completion of editing, alignments, and analyses of transcripts.
- Entering all primary data into secure database. Customization and integration of query analysis tools (OSU and University of Rijeka).
- In vivo studies. (University of Rijeka).
- Design of microarray probes.
- Development and purchase of MCMV gene arrays.
- Gene array experiments (OSU Microarray Core Facility).
- Preparation and submission of the first manuscript detailing MCMV transcriptome.
Year 3
- Completion of in vivo studies (University of Rijeka).
- Completion of gene array studies and confirmation of viral gene expression profiles
- Completion of databases with full integration of query software and bioinformatics tools.
- Completion of analysis of gene array data (OSU and University of Rijeka).
- Completion of MCMV transcriptome database.
- Preparation and submission of manuscripts.
