A Transcriptomic Approach to Viral Disease Research

Activities

 

Year 1

  1. Generation of three cDNA libraries from infected cells (representing three different temporal phases of infection).
  2. Performing positive selection for cDNA clones harbouring viral genes.
  3. Generation of glycerol stocks and plasmid DNA stocks for each viral cDNA clone (Parts a, b and c will be performed at the University of Rijeka).
  4. Sequence analysis of viral clones at OSU.  The rationale for this being done at OSU is that it can interact face-to-face with the sequencing facility to advance the specific needs and rapidly re-submit samples for sequencing at little or no cost.
  5. Database construction.
  6. Microarray platform selection.

Year 2

  1. Completion of sequencing of cDNA libraries generated in year 1 at OSU.
  2. Completion of editing, alignments, and analyses of transcripts.
  3. Entering all primary data into secure database. Customization and integration of query analysis tools (OSU and University of Rijeka).
  4. In vivo studies. (University of Rijeka).
  5. Design of microarray probes.
  6. Development and purchase of MCMV gene arrays.
  7. Gene array experiments (OSU Microarray Core Facility).
  8. Preparation and submission of the first manuscript detailing MCMV transcriptome.

 

Year 3

  1. Completion of in vivo studies (University of Rijeka).
  2. Completion of gene array studies and confirmation of viral gene expression profiles
  3. Completion of databases with full integration of query software and bioinformatics tools.
  4. Completion of analysis of gene array data (OSU and University of Rijeka).
  5. Completion of MCMV transcriptome database.
  6. Preparation and submission of manuscripts.
